Science and research

RESEARCH AREAS: anthropology, cell and developmental biology, tissue engineering, experimental morphology and oncology, clinical anatomy, and neuroscience.

METHODS USED: micro preparation, microsurgical techniques, histology and light microscopy, enzyme histochemistry, immunohistochemistry, lectin histochemistry, molecular biology techniques including in situ hybridization, tissue culture, fluorescence microscopy, optical mapping, and basic biochemical methods (ELISA, Western Blot). We use also computer modeling.

LABORATORY SPACE: The labs are located on the premises of the Institute of Anatomy. Additional lab space is found in the BIOCEV in Vestec (collaborative venture between the Charles University and Czech Academy of Sciences). More complex techniqes are performed in collaboration with the Czech Academy of Sciences as well as other national and foreign labs from countries including Germany, USA, France, South Korea, and Singapore.

Details of current research interests:

Studying the skull formation from the perspective of comparative and developmental anatomy using the collections of the Comparative Anatomy Museum of the Institute. Study of intracranial structures, malformations of the facial skeleton and skull base; modeling the soft tissue based on bony landmarks using anthropology approaches combined with CT and CBCT. Study of age-related changes of the face with the aim of identifying people based on skeleton or photographs. Reconstruction of selected specimens from our museum.

Study of molecular mechanisms that influence transformation of neural crest cells; characterization of stem cells of neural crest origin derived from human hair follicles using the chick embryo model and melanoma.

Additional research themes include phenotypic characterization of cells in squamous and glandular epithelia under normal and pathological conditions (basalioma, spinous cells carcinoma of the skin and ductal carcinoma of the pancreas). Epidermal stem cells and epithelia-mesenchymal interactions are studied under both normal conditions as well as wound healing or cancer growth.

Heart development and pathogenesis of heart failure are studied under normal and altered hemodynamic conditions with the aim to elucidate molecular mechanisms governing myocyte proliferation, conduction system differentiation, and factors modulating myocardial fibrosis.

Clinico-anatomical studies are currently focused on development of joints ligaments of the limbs in relation to growth-dependent changes on the skeleton.

Neuroscience research is focused on volumetric studies of the basal ganglia correlating gross anatomy with MRI in patients with Parkinson’s or Alzheimer’s disease, and those after carotid arteries occlusion.

Anthropology and experimental morphology:

Facial reconstruction based on the skull and subcutaneous tissue with a particular accent on main identifying features. Study of morphology of the upper airways with a focus on the vocal cords. The aim is to differentiate, based upon the architecture and tissue density, the individual components of the vocal apparatus and then digitally reconstruct human larynx as a template for individually tailored prosthetic replacements.

This project is a collaboration between our 3D printing facility and plastination lab and the Institute of Clinical and Experimental Medicine, Institute of Macromolecular Chemistry of the Czech Academy of Sciences and the Faculty of Mechanics and Mechanotronics of the Czech Technical University in Prague.

Cell Biology:

This laboratory studies the epithelial tissues under normal and pathological conditions. We study the interactions between the cancerous and non-cancerous cells in tumor microenvironment. Particular attention is paid to cancer-associated fibroblasts (CAFs) and their products. We focus on factors promoting the inflammatory response (IL-6, IL-8, and TGF-B). These factors appear to be the fundamental “words” in intracellular communication that influence tumor growth and metastasis. Considering the importance of inflammation in healing, tumor microenvironment and fatal viral infections we are looking for common junction points with the goal to better characterize them for ultimate therapeutic modulation. Lab activities are supported by the projects Center of Tumor Ecology and National Institute for Cancer research (funded by the EU).

Development of cardiovascular system:

We are currently studying the regenerative potential of reptilian heart, development of the cardiac conduction system, and morphogenesis and plasticity of the pectinate muscles in the vertebrate atria. We study the differential response of the adult heart to pressure and volume overload with particular attention to cardiac fibrosis in collaboration with the Institute of Clinical and Experimental Medicine. We are also developing and optimizing new techniques for imaging of the cardiovascular system. This research is supported by the grants of the Czech Science Foundation, Agency for Health-related research, and the Ministry of Education, Youth and Sports.

Clinical Anatomy:

Clinic-anatomical studies are focused predominantly on the ligaments of the large joints of the limbs and their relationship to growth-related skeletal changes, as well as growth and development of the cervical spine and possibilities of its osteosynthesis. We are involved also in pioneering selected surgical approaches with the aim to maximally preserve neurovascular structures, both open surgery and ultrasound-guieded procedures. In collaboration with our urogynecology colleagues we are optimizing application of mechanical devices supporting the urethra to restore continence.

Laboratory of neuroanatomy:

We continue our long-term collaboration with the Neurology and Radiology departments of the General University Hospital in the area of sleep medicine. We perform volumetric studies of the hippocampus and amygdala using ScanView software on MRI pictures, comparing patients suffering from narcolepsy and cataplexy with healthy individuals. In collaboration with the neurosurgeons from the Central Military Hospital we are studying the course of peripheral nerves on fixed cadavers and device the best surgical approaches. Further collaboration involves the hospital in Usti nad Labem in the area of teaching and clinically-oriented papers.
